What differences are there between these two apart from the colour of the wheels and the fact that the Plarail one has a coupling hidden in its nose?

I have had several pass through my hands but never noticed a difference. Of course I wasn't looking for any. Except what you mentioned, the yellow wheels and the retractable coupling, I didn't notice anything different but I didn't study them.

The JR East Co. Tsubasa Logo stick on the car is missing the hyper city version.
